Monacolin K's discovery:

Although the use of red yeast rice in food and medicine in China has been more than 1,000 years, it was not until 1979 that the efficacy of red yeast rice attracted attention.

In 1979, Professor Akira Endo of Japan isolated an active substance from Monacolus monascus and named it "Monacolin K", which attracted the attention of the food and pharmaceutical community to Monacolin K.

Later, United States geneticists Brown and Goldstein confirmed the mechanism of action of Monacolin K to inhibit cholesterol synthesis, for which they were awarded the Nobel Prize in 1985. Since then, a new type of red yeast rice "functional red yeast" has been officially born.

It has now been proposed that the real lipid-lowering substance in red yeast rice, Mevi-nolinic A cid (known as "acid monepectolin K"), can directly exert lipid-lowering effects without the participation of hydroxyl esterase in the human body. Lovastatin is a prescription drug developed by Merck in United States, which is hexahydronaphthalene ester, a prodrug with a lactone structure. It is inactive by itself, and needs to be hydrolyzed by hydroxyesterase produced by the human body to change its structure before it can be active and exert lipid-lowering effects. The molecular formula, molecular structure, and molecular weight of the two are completely different. It was found that this acid-style monepolin K was only found in a small number of naturally fermented red yeast rice.


From a scientific point of view, they are two substances with different origins, different structures, and different properties. Since Lovastatin is a prodrug and has no lipid-lowering effect, it needs to be hydrolyzed by the hydroxysterase produced by the human body to exert its efficacy. However, there are at least three types of hydroxysterase produced by the human body, and there are large individual differences in the ability to produce hydroxysterase, and some people even have none at all. This determines that the lipid-lowering effect of lovastatin varies greatly in different patients. Patients with more hydroxyesterase have a good lipid-lowering effect, while patients with less hydroxysterase have a poor lipid-lowering effect and have great toxic side effects. In some cases, it is completely ineffective.


The lipid-lowering substances in the naturally fermented red yeast rice, because most of them are already in the acid structure, that is, the acid M onacolin K, no longer need the hydroxyesterase in the human body to participate in hydrolysis, so it does not increase the burden on the liver and kidneys, and directly plays a lipid-lowering effect, therefore, the lipid-lowering effect is better than that of Lovastatin, and the toxic side effects are less than that of Lovastatin.

The effect of monacolin K on the human body

1. Hypolipidemic effect

A large number of clinical trials have shown that the Mon acolin substances discovered by Professor Endo are effective, low-toxicity and effective drugs for the treatment of hypercholesterolemia, which can strongly reduce the cholesterol content in the body, and have a specific inhibitory effect on the rate-limiting enzyme hydroxymethylglutaryl-CoA (HMG-CoA) reductase, which is the rate-limiting enzyme for the synthesis of cholesterol, and the concentration of such substances reaches 0.001~0.005Hg/mL, and the synthesis of cholesterol will be significantly hindered [3-11].

Professor Endo believes that the content of Monacolin K in the human blood reaches 1-5Ag/L, which has the effect of lowering cholesterol, while the blood volume of adults is 20L, and the effective dose of Monacolin K required is 100Hg, and only 10~30 mg of Monacolin K can significantly reduce the cholesterol content every day [123]. The conventional treatment dose can reduce the total cholesterol (TC) in the plasma by 15%~20%, LDL-C (low-density lipoprotein cholesterol) by 10%~40%, and increase HDL-C (high-density lipoprotein cholesterol) by 2%~15%, and the side effects are few and mild.


2 Anti-atherosclerotic (AS) effect.

AS is one of the most important diseases endangering human health, and its pathophysiological changes are mainly the proliferation of VSMCs (vascular smooth muscle cells), internal and inferior activity and lipid metabolism disorders. Hyperlipidemia is the main cause of causing and exacerbating AS. Human blood contains a lot of fats, such as cholesterol, triglycerides, free fatty acids, etc., which are often referred to as blood lipids. With the increase of age, the blood lipid content will gradually increase, and the cholesterol (CH) in it can break through the barrier of the vascular endothelium, so that its permeability increases, and plasma lipoproteins can enter the vascular intima to cause the clearance response of macrophages and the proliferation of smooth muscle cells in the blood vessel wall to form AS plaques.

At the same time, a large amount of CH, LDL, and VLDL will also be gradually deposited on the inner wall of blood vessels, forming "blood scale", and over time, "blood scale" will be deposited more and more in blood vessels, forming atherosclerotic plaques, and finally blocking blood vessels, resulting in cerebral thrombosis, stroke, coronary heart disease, arteriosclerosis, myocardial infarction and other cardiovascular and cerebrovascular diseases. Atherosclerotic lesions occur in almost everyone, but the time of occurrence is early and late, the degree is severe, the location is different, and the development is fast and slow.

3 Inhibition of restenosis after transluminal coronary angioplasty (PTCA).

Restenosis is the main factor affecting the efficacy of PTCA, and the incidence rate is 25%~40% 3~6 months after surgery. The mechanism of restenosis is mainly due to the migration and proliferation of vascular smooth muscle cells (VSMCs) in the middle layer to the intima after intravascular injury, resulting in intimal thickening, of which the proliferation of VSMCs is the main factor. Recent studies have found that the HMG-R inhibitor MonacolinK can inhibit the proliferation of VSMCs, thereby reducing the incidence of restenosis after PTCA.


Increase the content of monacolin K in red yeast rice

Monacolin compounds in the fermentation products of Monascus have a significant role in regulating blood lipids, especially Monacolin K is considered to be the best blood lipid regulator at present. However, it has been found that the content of Monacolin K in red yeast rice is very low, only a few parts per million, so people have used various methods to increase the content of Mona-colin K in red yeast rice. There are two main ways to start from it: one is to screen for high-yielding strains, and the other is to change the growth conditions to increase its yield.
